I love most if not all newhus and will always support those pushing content for them, as well as creators looking for characters to use.
In the realm of Stage 5 subordinates, Mayumi stands out as an active leader & user of an army of Haniwa soldiers, in a sea of characters that mostly fight as their master's last stand (Orin has the zombie fairies, sort of?)

Mayumi, Saki, Biten are three that seem almost as fightan plants. The rest of Animal Hell can be more or less reasonably put into such game as well, I think.
Get some justification for UDoALG girls (Biten, Chiyari, Enoko) to get back into yakuza business, three leaders, Mayumi and Keiki, round it out with Youmu and Tenshi as Gensokyo representatives and you have 10 characters for a fangame fightan, almost as much as what IaMP was. Is it hard to produce one? I'm sure it's harder than making shmup, because of balance and such, but what is a standard amount of work put into fighting game?

I wonder if you couldn't slot Youmu and Tenshi out for Ran and maybe Megumu. It fits with the animal theme of the yakuza, are characters who haven't had a fighting entry, and are both in positions to have some authority and ulterior motives behind them. Ran could be the boring route where she does her best to maintain Yukari's status quo and takes her traditional role in these games, but at the same time you can give her some side plot where she bends the rules a bit and throw her in with Yuuma. Not sure if I'd do another AoCF with a tag team system, but they'd make a good team I think. Megumu could be working to advance the strength of the Tengu and maybe undermine Moriya's influence over them in working with the yakuza. She's no stranger to shady dealings and you've got a criminal underbelly trying to shake things up, and following Chimata's incident the game could resolve that in some interesting ways.

I've proposed Youmu because she could fulfill the role of some familiar Touhou protagonist character and at the same time, as Youmu is tied to Ministry of Right and Wrong, you can reasonably see her involved in something from Animal Hell without whole Gensokyo (thus Marisa and Reimu) getting dragged in.
Tenshi on the other hand is a loose canon that could get involved just for the fun of it, has the history of being fightan-hu and offers funny contrast (celestial in hell).
You are spot on with Ran, I can't believe I forgot about her. I'm not so sure on Megumu, although there is an appeal to seeing her getting physical with her tripod.
I thought now about Kutaka as another Ministry oriented 2hu to go along with Youmu, which would give two characters per "faction": each gang's leader and subordinate, Mayumi and Keiki, Youmu and Kutaka for Ministry, Ran and someone else for unaffiliated Gensokyo.
